The protocol of Ulcerative Colitis Mice Modeling using Dextran sodium sulfate (DSS)

The mouse model is the most common model for colitis modeling. After the hamster ulcerative colitis model was first prepared by using dextran sulfate sodium (DSS) in 1985, there are a lot of data to prove the etiology and clinical characteristics of the DSS colitis model. Symptoms, pathological changes, and treatment responses are similar to those of human ulcerative colitis (UC) [1].

1. Protocol

1.1 Acute modeling

1) Sterile drinking water with 3%-5% DSS, and filter with a 0.22 μm membrane, the solution must be colorless or slightly yellowish and clear.
2) Mice are housed in groups, at least 6 mice per cage, and the animals are maintained under standard conditions (with a temperature of 25℃, a 12-h/12-h light/dark cycle, and ad libitum access to food and water)
3) The body weight, diarrhea, and blood in the stool of the mice were recorded every 24 hours.
4) After rearing for a week, the DSS aqueous solution was replaced with an ordinary DSS-free aqueous solution, and the index was finally recorded [2].

1.2 Chronic modeling

1) Prepare 1%-3% DSS sterile aqueous solution, and the operation is the same as that of acute modeling.
2) Same as acute modeling 2)

4) Drink freely for several weeks, and finally replace the DSS aqueous solution with an ordinary DSS-free aqueous solution, and finally record the indicators [3].

3. Successful case for modeling with Yeasen DSS

Yeasen provides high-quality DSS (Cat#60316ES): purity (98%), sulfur content 17-19%, free sulfur <0.2%, supported by a large number of literature data.
